چکیده

SUMMARY Urethan, a hepatocarcinogen in adult regenerating liver, was administered to rats at various intervals after partial hepatectomy to determine the effect of this drug on (a) the pattern of DNA replication and (b) the induction by partial hepatectomy of some enzymes required for DNA synthesis. A single i.p. dose of urethan, 1 mg/g, was injected into rats at either 0.5, 12, or 24 hr after surgery. When urethan was given 0.5 hr after partial hepatectomy, the initiation of DNA synthesis was delayed by 6 hr, followed by greater uptake of thymidine-3H into nuclear DNA than into that of untreated regenerating liver. When urethan was given 12 hr after partial hepatectomy, the first wave of DNA synthesis was suppressed by 50 to 60%; the second cycle of DNA replication was essentially unaffected. Administration of urethan 24 hr postsurgery resulted in maximum inhibition at 14 hr after treatment; the incorporation of thymidine-3H during the second wave of DNA replication occurred within a shorter period of time than that of the control. The induction of certain enzymes linked to DNA replication during liver regeneration was affected by the urethan in a similar manner. Ribonucleoside diphosphate reductase and thymidine kinase were sensitive to urethan treatment, while DNA polymerase activity was insensitive. Other enzymes not tightly coupled to DNA synthesis in regenerating liver, such as adenine and hypoxanthine-guanine phosphoribosyl transferases, were not affected by the carcinogen. The results indicate that urethan selectively inhibits the induc tion by partial hepatectomy of enzymes necessary for DNA synthesis.

Effects of 6-thioguanine on macromolecular events in regenerating rat liver.

Regenerating rat liver was used as a semisynchronous system in which to investigate the effects of 6-thioguanine on biochemical processes occurring in discrete phases of the cell cycle. 6-Thioguanine inhibited the first wave of DNA biosynthesis in regenerating rat liver. This effect appeared to be the result of a decrease, caused by 6-thioguanine, in the induction of several enzyme activities (...

Gnidilatimonoein from Daphne mucronata inhibits DNA synthesis in human cancer cell lines

The anticancer agents from plant sources usually exert their action through a wide range of mechanisms. As part of our studies of plants for new anticancer agents with emphasis on Thymelaeaceae family, we examined the cytotoxicity and anti-tumor activity of the water extract of D. mucronata leaves against induced breast tumor in rats. In the current study, we were interested to obtain some know...

Inhibiting effect of caffeine on spontaneous and urethan-induced lung tumors in strain A mice.

The i.p. injection of caffeine (8, 20, and 40 mg/kg) 3 times weekly for 8 weeks suppressed the development of spontaneous pulmonary adenomas in strain A mice. The same caffeine injection scheme suppressed urethan (0.25 and 1.0 mg/g)-induced lung tumor development when caffeine treatment started 1 week before urethan administration, but this suppression was not significant when caffeine treatmen...

Studies on the biosynthesis of deoxyribonucleic acid by soluble mammalian enzymes.

A variety of reactions associated with the anabolism of ribonucleotides and deoxyribonucleotides as well as with their incorporation into ribonucleic acid and deoxyribonucleic acid, respectively, has been shown to occur in the high speed, particle free, supernatant fraction of rat liver. Such a cellular fraction obtained from normal rat liver has been shown to convert UMP (I), dAMP, dCMP, and d...

[Methyl-14C]-choline incorporation into nude mice bearing tumor xenografts-correlation with [methyl-3H]-thymidine: A pilot study

Background: Many in vitro studies suggested that choline incorporation into many types of tumors is related to cell proliferation. Whether in vivo choline incorporation is related also to cell proliferation or not was my question. Therefore, the aim of this pilot study was to investigate the relationship of in vivo [methyl- 14C]-choline incorporation and in vivo [methyl-3H]- thymidine incorpora...
